Calcium mobilization from mitochondria in synaptic transmitter release
نویسنده
چکیده
Mitochondria can rapidly accumulate and release Ca2+ upon cell stimulation. A paper by Yang and coworkers in this issue reports an unusual form of synaptic potentiation, dependent on Ca2+ release from mitochondria through the Na+/Ca2+ exchanger and triggered by Na+ entry through voltage-gated channels (Yang et al., 2003).

Primary and secondary regulation of quantal transmitter release: calcium and sodium.
Calcium is the prime regulator of quantal acetylcholine liberation at the neuromuscular junction; its entry through the presynaptic membrane and the level of free [Ca]in most probably determine the number of transmitter quanta liberated by the nerve impulse. The level of free [Ca[in, in turn, is controlled by a number of subcellular elements: mitochondria, endoplasmic reticulum, vesicles, macro...
متن کاملAssessing the roles of presynaptic ryanodine receptors and adenosine receptors in caffeine-induced enhancement of hippocampal mossy fiber transmission.
Caffeine robustly enhances transmitter release from the hippocampal mossy fiber terminals, although it remains uncertain whether calcium mobilization through presynaptic ryanodine receptors mediates this enhancement. In this study, we adopted a selective adenosine A1 blocker to assess relative contribution of A1 receptors and ryanodine receptors in caffeine-induced synaptic enhancement. Applica...
متن کاملPresynaptic mitochondrial calcium sequestration influences transmission at mammalian central synapses.
Beyond their role in generating ATP, mitochondria have a high capacity to sequester calcium. The interdependence of these functions and limited access to presynaptic compartments makes it difficult to assess the role of sequestration in synaptic transmission. We addressed this important question using the calyx of Held as a model glutamatergic synapse by combining patch-clamp with a novel mitoc...
متن کاملCalcium Stores in Hippocampal Synaptic Boutons Mediate Short-Term Plasticity, Store-Operated Ca2+ Entry, and Spontaneous Transmitter Release
Evoked transmitter release depends upon calcium influx into synaptic boutons, but mechanisms regulating bouton calcium levels and spontaneous transmitter release are obscure. To understand these processes better, we monitored calcium transients in axons and presynaptic terminals of pyramidal neurons in hippocampal slice cultures. Action potentials reliably evoke calcium transients in axons and ...
متن کاملPresynaptic calcium diffusion from various arrays of single channels. Implications for transmitter release and synaptic facilitation.
A one-dimensional model of presynaptic calcium diffusion away from the membrane, with cytoplasmic binding, extrusion by a surface pump, and influx during action potentials, can account for the rapid decay of phasic transmitter release and the slower decay of synaptic facilitation following one spike, as well as the very slow decline in total free calcium observed experimentally. However, simula...
